CRYSTAL VIOLET

Crystal Violet 0.5gms
Distilled Water 100mls

  1. Dissolve the crystal violet in alcohol before adding any water (10mls of alcohol per gram of crystal violet)
  2. When dissolved add the water
  3. Heat to 80oC on a stirrer to evaporate the alcohol
  4. Filter

GRAM'S IODINE

Iodine 1gm
Potassium Iodine 2gms
Distilled Water 300mls

  1. Dissolve the Potassium Iodine in a little water (1.66mls of distilled water per gram of potassium iodine)
  2. Dissolve iodine in this solution
  3. Make up with the remaining water
  4. DO NOT FILTER

SAFRANIN

Safranin 0.5gms
Distilled Water 100mls

  1. Dissolve Safranin in the water
  2. Leave for one half to a whole day on the magnetic stirrer
  3. Filter

ALBERT'S STAIN

Toluidine Blue 0.15gms
Malachite Green 0.2gms
95%Ethanol 2mls
Glacial Acetic Acid 1ml
Distilled Water 100mls

  1. Dissolve the dyes in alcohol
  2. Add the distilled water and the acid
  3. Filter

MALACHITE GREEN

Malachite Green 5gms
Distilled Water 100mls

  1. Dissolve Malachite Green in water
  2. Leave for one half to a whole day on the magnetic stirrer
  3. Filter

METHYLENE BLUE

Methylene Blue 0.5gms
Distilled Water 100mls

  1. Dissolve Methylene Blue in water
  2. Leave for one half to a whole day on the magnetic stirrer
  3. Filter

ZN CARBOL FUCHSIN

Powdered Basic Fuchsin 5gms
Phenol 25gms
95% Alcohol 50mls
Distilled Water 500mls

  1. Dissolve the Fuchsin in Phenol with a little water over a boiling waterbath
  2. Add the alcohol and mix
  3. Add the rest of the water
  4. Filter

The deposit which develops when the alcohol is added will dissolve when the water is added and the stain placed on the stirrer for at least half a day

LACTOPHENOL COTTON BLUE

Phenol 20gms
Glycerol 40gms
Lactic Acid 20gms
Cotton blue 0.05gms
Distilled Water 20mls

  1. Weigh a beaker with a 250ml capacity
  2. Weigh phenol and glycerol into the beaker
  3. Swirl to dissolve (It should only take approx. 5 mins)
  4. Weigh in lactic acid
  5. Dissolve the cotton blue, (also known as methyl blue) in the water in a separate beaker
  6. Leave overnight to eliminate insoluble dye
  7. Mix the two solutions together

DILUTE CARBOL FUCHSIN

Carbol Fuchsin 0.5gms
10% Ethanol 100mls

(10% Ethanol = 10.53mls of 95% Ethanol + 89.47mls of Distilled Water)

  1. Dissolve Carbol Fuchsin in 95% Ethanol
  2. Put on magnetic stirrer for an hour or so (not all will dissolve)
  3. Add distilled water
  4. Filter

QUINK STAIN

10% Potassium hydroxide 70mls
Glycerol 40mls
Parker 'Quink' 10mls
(Permanent blue or blue/black)

  1. Mix together
  2. Does not require filtering
