Reagents

CATALASE REAGENT

3% H2O2, prepared as follows:
H2O2 stock solution, 30% w/v 10ml
MQ water 90ml

Store at 4oC in a light proof bottle.

10% FERRIC CHLORIDE (TDA)

Ferric Chloride (anhydrous) 10gms
Distilled water to 100mls Weight/Volume

(For FeCl3.6H2O you require 16.663gms per 100mls. This can be used instead of Ferric Chloride (anhydrous))

KOVACK’S REAGENT

Para Di Methyl amino benzaldehyde 5gms
n - Amyl alcohol (n - pentanol) 75mls
Hydrochloric acid (SpG 1.19) 25mls

  1. Heat alcohol and aldehyde in water bath at 50 - 60 oC until dissolved
  2. Cool and slowly add the HCl
  3. Store at 4oC

NITRATE REDUCTASE REAGENTS:

0.1% NAPHTHYL ETHYLENE DIAMINE DiHCl (NITRATE A)

Naphthyl ethylene diamine di HCl 0.1gm
Distilled water 100mls

  1. Dissolve naphthyl ethylene diamine diHCl in water
  2. Store in dark bottles at 4oC

0.2% SULPHANILAMIDE (NITRATE B)

Sulphanilamide 0.2 gms
Distilled water 100mls

  1. Dissolve sulphanilamide in the water
  2. Use the magnetic stirrer - It may take hours!
  3. Store at 4oC

OXIDASE STRIPS

N,N,N’,N’, tetramethyl-p-phenylenediamine, made up as a 1% w/v aqueous solution, is used to soak filter paper strips (approximately 1x3 cm) which are then dried and stored in a dark container. The reagent is subject to auto-oxidation and colour change if exposed to light.

VP REAGENTS:

40% POTASSIUM HYDROXIDE (VP1)

KOH 40gms
Creatine 0.3gms
Distilled water to 100mls Weight/Volume

  1. Dissolve the KOH in most of the water
  2. Dissolve the Creatine in the remaining water
  3. Add the two solutions together volumetrically
  4. Store at 4oC



ALPHA – NAPHTHOL (VP 2)

Alpha - naphthol 6gms
95% Alcohol to 100mls W/V

  1. Dissolve alpha - naphthol in alcohol
  2. Store in dark bottles at 4oC
